An Experimental Study on Histological Changes in the Gonads, and Changes in Content of Water and Electrolytes in the Tissue of Uterus and Lung of Mature Female Rabbits by Resepine Adrministration


Abstract
Reserpine, one of the tranquilizers and rauwolfian alkaloids, has been widely used as a anti-hypertensive drug in the obstetrical field. There has been some reports that the reserpine decreases blood pressure by reducing the heart beat rate because reserpine decreases or consumes catecholamine in the cardiac tissue. Although the drug decreases blood pressure, it has a number of untoward side effects affecting various other organs. In addition, reserpine disturbes ovulation and implantation of the fertilized ovum and causes atrophy of the uterus and the ovaries by influencing hypothalamuspituitary-gonadal or hypothalamus-pituitary-adrenal axis.
An attempt was made to clarify the ovarian changes by reserpine and its effect on water and electrolytes in the uterine and lung tissue of mature female rabbits weighing around 2kg of body weight.
A group of rabbits preconditioned with 50 I. U/kg of HCG and another with 1.5mg/kg of reserpine plus 50 I. U/kg of HCG were observed in 24 hours after conditioning.
The results obtained are as follows:
1. The HCG preconditioned mature female rabbits showed an acceleration of maturation of the follicles and ovulation in the ovaries, and reserpine plus HCG preconditioned group showed inhibition of the accelearating maturation in ovaries that was caused by HCG preconditioned and also showed corpus luteum formation and cystic change of the corpus luteum.
2. The electrolyte levels in the serum of HCG preconditioned rabbits showed decrease in chloride and magnesium and increase in sodium, potassium and calcium levels. The level of chloride was in-creased and potassium, calcium and sodium levels decreased furthermore. Therefore, it was evidenced that the reserpine inhibited the electrolyte change& caused by HCG preconditioning.
The water, sodium and calcium levels in the uterine tissue of the HCG preconditioned group showed increase in the contents compared to the normal group and decrease in chloride and magnesium. In reserpine plus HCG preconditioned group, level of water was increased and showed further decreased levels in chloride, potassium, and magnesium. The water and the electrolyte changes induced by the HCG preconditioning could not be inhibited by the reserpine.
The contents of chloride and potassium in the lung tissue of the HCG preconditioned group were increased whereas sodium, calcium, and magnesium contents were decreased. The water, sodium, and magnesium contents were increased, and chloride and potassium contents were decreased in reserpine plus HCG preconditioned group. Although the reserpine showed the inhibitory action to water and electrolytes in the lung tissue, it did not influence on the calcium contents.
The water contents in the lung tissue of the normal, HCG and reserpine plus HCG preconditioned group were lower than those of the uterine tissue.
The chloride contents were higher in the lung tissue of the normal, HCG, and reserpine plus HCG preconditioned group compared them to the uterine tissue level.
The sodium contents in the lung tissue of the normal was higher and HCG preconditioned group showed higher level in the uterine tissue. Its content in the lung tissue of reserpine plus HCG preconditioned group was increased again.
While the potassium contents in lung and uterine tissue of normal showed no significant difference its levels were increased in lung tissue in HCG and reserpine plus HCG preconditioned group. The calcium contents in lung tissue of the normal, HCG, and reserpine plus HCG preconditioned group were higher than those of the uterine tissue.
0. The magnesium contents in the lung tissue of the normal showed decreased while the HCG preconditioned group showed no appreciable changes. Its content in the lung tissue of reserpine plus HCG preconditioned group showed increased level in comparison to its level in the uterine tissue.
